Why Multiplexing

With the growth of precision medicine and the development of personalized medicine, the number of available assays has been increasing along with the cost of healthcare. Multiplexing provides an attractive solution for cost-reduction, as unlike traditional methods, a multiplex assay can provide results for multiple targets in a single reaction, saving time, labor, reagents, samples, and thereby reducing healthcare costs.

 

 

What is πCode™ MicroDisc?

πCode is acronym for “Precision Image Code”, it is a circular disc manufactured by semi-conductor processes and able to generate over 16,000 distinct image patterns for multiplexing applications. To be used in a broad spectrum of applications, each circular microdisc is encapsulated in a highly stable matter with an added paramagnetic property, which allows it to be suitable for conjugation, washing, and automation procedures.

 

How does it work?

Each πCode MicroDisc with a distinct circular image pattern corresponds to the specific capture agent conjugated to the disc surface.  This capture agent allows the capture and detection of targets from a sample. Virtually any probe used in clinical diagnostics can be conjugated to πCode MicroDiscs, including DNA, RNA. antigens, antibodies, proteins, or chemical compounds.

 

 

In order to perform a biological test, πCode MicroDiscs with desired capture probes were pooled together in a single well and reacted with patient sample.  For example, there are 40 mutations/targets to be detected in our IntelliPlex™ EGFR Mutation Kit, hence, the number of different patterns of πCode MicroDiscs present in the assay is 40 types.  The assay then is followed by hybridization, fluorescent labeling, and detected by optical imaging fluorescence analyzer PlexBio™ 100 - which uses the CCD camera to read the distinct image patterns under bright field and quantifies the target under dark field.  PlexBio™ 100 Fluorescent Analyzer measures the fluorescence signal intensity (MFI) releases by each target and provides a comprehensive report in a user-friendly interface.
